Is there a possibility of using calculate function with matrix questions? I have tried using the label for each row/column of the matrix but it is not recognised when I try use the preview function.

I think you need to use variable names, not the labels.

You could use calculate within the matrix question type as outlined in the image below:
In the survey tab of your xlsform:
In the choices tab of your xlsform:
Data entry screen as seen in Enketo: The values within the red highlighted box is an auto calculated value.
